Storage and Stability

Alhydrogel 2% is shipped at room temperature and should be stored at room temperature. The product is stable for 6 months. DO NOT FREEZE.

Description

Alhydrogel is a wet gel suspension of aluminum hydroxide (called alum). Alum improves the attraction and absorption of antigen by antigen-presenting cells (APC). Recently, it has been suggested that antigens adsorbed on aluminum salts are presented in the form of particles, which internalize them more efficiently by APCs.

In addition, alum activates the NLRP3 inflammasome complex involved in the induction of several pro-inflammatory cytokines, including IL-1β and IL-18. Alum increases Th2 antibodies but does not promote a significant Th1 cellular response. 2% hydrogel was chosen as the international standard preparation for aluminum hydroxide gels. Alhydrogel 2% is present in multiple commercial vaccine formulations.

Preparation of antigen-2% hydrogel mixture

The antigens are preferably diluted in saline or phosphate buffers. The amount of protein or peptide conjugate used for primary immunization can be adjusted depending on the availability and immunogenicity of the antigen. For example, mice can be injected subcutaneously (s.c.) with 1 to 10 μg of endotoxin-free ovalbumin.

1. Shake the capped bottle of Alhydrogel 2% well before use.
2. Add Alhydrogel 2% to the antigen solution; The final volume ratio of Alhydrogel 2% to antigen should be 1: 1 (100 μl Alhydrogel 2% per 100 μl antigen) to 1: 9 (100 μl Alhydrogel 2% per 900 μl antigen).
3. Mix well by pipetting up and down for at least 5 minutes to allow Alhydrogel 2% to effectively adsorb the antigen.
